Preparation of ES Cells for Karyotyping

  1. Notify the Transgenic Mouse/ES Cell Shared Resource that you have cells that you would like to karyotype. The Tg Mouse/ESC Shared Resource will schedule and coordinate a time for cells to be delivered for karyotyping.
  2. E-mail the names of the cell lines requested for karyotyping to Jake Stein (jake.stein@vanderbilt.edu) and Jennifer Skelton (jennifer.skelton@vanderbilt.edu). Label the culture dishes with the same names.
  3. On a 60 mm culture plate, plate cells on a layer of MEFs.
  4. Colonies need to be at least medium in size and 50% to 90% confluent for karyotyping. This is usually about two days of growth.
  5. On the day dishes are being handed off to the Shared Resource for karyotyping, feed the plate(s) before 10 a.m. and communicate to the core personnel what time they were fed.
  6. Bring plates for karyotyping to Shared Resource (9410 MRB IV) by 11:30 a.m.
