Cre or Flpe Pronuclear DNA Microinjection

The resource performs these microinjections directly into fertilized embryos derived from gene-targeted mice, but males studs need to be provided by the investigator. Utilizing a supercoiled Cre or Flpe expression plasmid, removal of "floxed" or "flrted" gene segments of DNA from the genome can be achieved. This charge for this service is $4,800. Currently, three core employees can perform Cre or Flpe pronuclear DNA microinjections.

The investigator will be notified of potential transgenic pups as weaning of these pups nears. Pups will be tailed and numbered at 3 weeks of age by core personnel. The investigator will need to perform DNA isolation and analyze the DNA by PCR and Southern blotting. If no transgenic models are generated, a decision will need to be made as to the concentration of the Cre or Flpe being injected.
