Freezing sperm is more cost effective and less labor intensive than freezing 8-cell embryos. However, reconstitution of the line by in vitro fertilization is not as easy to accomplish as transfer of embryos into the female reproductive tract. Sperm viability from outbred strains of mice is adequate, however, viability from inbred strains of mice can be less than satisfactory for a successful rederivation of a mouse line.
The use of this service is initiated after consultation with a member of our staff. The investigator is responsible for providing a complete description of the mouse line to be cryopreserved (strain, genotype, etc.). Upon completing the appropriate form, sperm is obtained from the epididymides and vas deferentia of a male transgenic or knock-out mouse and cryopreserved.
Vials of frozen sperm are stored in liquid nitrogen storage tanks. These tanks are monitored weekly by personnel of the resource, and cannot be accessed by other individuals. This service is available to investigators at $240 per male mouse. There is an additional service charge for long term storage. Currently, three core personnel are capable of performing this service.
